The X was an amplified DNA sample that contained pyrophosphate (PPi), which was released after nucleotide bases were incorporated into the DNA template. It was subsequently reacted with the enzyme inorganic pyrophosphatase (PPase) to transform it into inorganic phosphate (Pi). PiColorLock™ dye reagents was react with inorganic phosphate (Pi) to turn the color in green.
